白杨黄素对成骨细胞系MC3T3-E1成骨向分化能力的影响

Effect of anti-inflammatory flavonoid chrysin on osteogenesis of preosteoblast MC3T3-E1 cells

摘要: 目的: 探讨黄酮类小分子白杨黄素对成骨细胞系MC3T3-E1成骨向分化能力的影响。方法: 检测白杨黄素的细胞毒性后,确定最佳作用浓度。采用DMEM完全培养基将成骨细胞系MC3T3-E1制成单细胞悬液,加入浓度为25 μmol/L的白杨黄素作为实验组,以单纯DMEM组作为阴性对照组,以成骨诱导液组(普通DMEM培养基中加入10 mmol/L β甘油磷酸钠、50 μmol/L维生素C和10 nmol/L地塞米松)作为阳性对照组。体外培养后,运用实时定量PCR分别检测早、中、晚期成骨相关指标Runx2、ColA1和OCN mRNA的表达。采用SPSS17.0软件包对数据进行统计学分析。结果: 浓度为25 μmol/L的白杨黄素对MC3T3-E1细胞培养无毒性;在添加25 μmol/L的白杨黄素共培养7 d后,Runx2的mRNA表达量达到峰值, 14 d后ColA1的mRNA表达量达到峰值;共培养21 d后,OCN的mRNA表达量达到峰值。结论: 25 μmol/L的黄酮类小分子白杨黄素能够促进成骨细胞系MC3T3-E1成骨分化过程中成骨相关基因的表达。

Abstract: PURPOSE: To investigation the effect of flavonoid chrysin on osteogenesis of preosteoblast MC3T3-E1 cells. METHODS: After evaluation of the toxic effect of chrysin, preosteoblast MC3T3-E1 cell suspension was prepared with DMEM and then the cells were cultured with 0 and 25 μmol/L chrysin. Real-time quantitative PCR was used to detect the expression of osteogenic marlcers including Runx2, ColA1 and OCN. Statistical analysis was performed using SPSS17.0 software package. RESULTS: 25 μmol/L chrysin had no toxic effect on preosteoblast MC3T3-E1 cells. After co-culture with 25 μmol/L chrysin,the expression of Runx2, ColA1 and OCN was highest at the 7th,14th and 21th day,respectively.CONCLUSIONS: : 25 μmol/L chrysin can promote osteogenic gene expression of preosteoblast MC3T3-E1 cell.
