牙龈癌中VEGF及PTEN的表达及其相关性分析

Correlation analysis of VEGF and PTEN expression in gingival carcinoma

摘要: 目的 研究血管内皮细胞生长因子(vascular endothelial growth factor, VEGF)及张力蛋白同源基因(phosphatase and tensin homolog, PTEN)在牙龈癌中的表达及相关性,为临床治疗提供依据。方法 免疫组织化学S-P法研究66例牙龈癌(高分化31例,中分化20例,低分化15例)患者和15例癌旁正常组织的VEGF及PTEN的表达情况,应用SPSS13.0软件包分析2种因子的表达与临床特征之间的相关性。结果 癌旁正常组织中PTEN的阳性率高于其他各组,差异具有统计学意义(P<0.05)。低分化组中VEGF的阳性率最高,与其余组比较差异显著(P<0.05)。相关性研究结果显示,VEGF与复发和淋巴结转移呈正相关(P<0.05);PTEN与复发和淋巴结转移呈负相关(P<0.05);VEGF与PTEN在牙龈癌中的表达呈明显负相关(P<0.05)。结论 VEGF表达增加和PTEN表达降低可能在牙龈癌发生、发展过程中共同发生作用。

Abstract: PURPOSE: To study the expression of vascular endothelial growth factor (VEGF) and phosphatase and tensin homolog (PTEN) in gingival carcinoma and their correlation in order to provide reference for clinical treatment. METHODS: Sixty-six gingival cancer patients were determined by pathological examination, among which 31 were well-differentiated, 20 were moderately differentiated and 15 were poorly differentiated.15 adjacent normal tissues were chosen as control group. The expression of VEGF and PTEN was examined by immunohistochemical method. Correlation analysis was performed with SPSS13.0 software package. RESULTS: The positive rate of PTEN in control group was significantly higher than other groups (P<0.05). The expression of VEGF in poorly differentiated group was significantly higher than other groups (P<0.05). Correlation analysis showed that the expression of VEGF was positively correlated to recurrence and lymph node metastasis (P<0.05); PTEN was negatively correlated to recurrence and lymph node metastasis (P<0.05); VEGF and PTEN in gingival carcinoma was negatively correlated (P<0.05). CONCLUSIONS: The increased expression of VEGF and decreased expression of PTEN in gingival carcinoma may play a mutual role in the development of gingival cancer.
