GSTP1 Ile105Val基因多态性与口腔癌易感性的meta分析

GSTP1 Ile105Val polymorphism confer susceptibility to oral cancer: a meta-analysis

摘要: 目的:采用meta分析评价谷胱甘肽-S-转移酶P1(GSTP1)基因多态性与口腔癌易感性的关系。方法:计算机检索PubMed、CBM、万方、维普、CNKI等中英文数据库,检索范围从建库至2013年7月15日,全面收集相关文献数据并计算优势比(OR)及95%置信区间(CI),以评价其关联性。结果:共纳入13项病例对照研究,包括2019例口腔癌患者及3282例健康对照人群。Meta分析结果表明,总体而言,GSTP1 Ile105Val基因多态性与口腔癌易感性之间不存在显著相关性(Val vs. Ile: OR=1.02, 95%CI: 0.91-1.15, P=0.719; Val/Val+Ile/Val vs. Ile/Ile: OR=1.03, 95%CI: 0.92-1.16, P=0.572)。基于种族不同的亚组分析结果表明,GSTP1 Ile105Val基因多态性与口腔癌发病风险的关联性在东亚人群(2项纳入研究)(Val vs. Ile: OR=1.42, 95%CI:1.06-1.90, P=0.019; Val/Val+Ile/Val vs. Ile/Ile: OR=1.59, 95%CI: 1.13-2.23, P=0.007)和欧洲高加索人群(2项纳入研究)(Val vs. Ile: OR=1.34, 95%CI: 1.03-1.75, P=0.032; Val/Val+Ile/Val vs. Ile/Ile: OR=1.66, 95%CI: 1.15-2.38, P=0.006)中存在显著关联性,但这一关联性在南亚人群(4项纳入研究)与美洲高加索人群(5项纳入研究)中并不显著。结论:现有证据表明,GSTP1 Ile105Val基因多态性与口腔癌易感性在东亚人群与欧洲高加索人群中存在显著关联。然而,GSTP1 Ile105Val基因多态性与口腔癌易感性之间的关联性,有待更大样本的病例对照研究加以证实。

Abstract: PURPOSE: This meta-analysis was aimed to evaluate the association of glutathione S-transferase P1 (GSTP1) Ile105Val genetic polymorphism with susceptibility to oral cancer. METHODS: A literature search of PubMed, Chinese BioMedical, Wanfang, VIP, CNKI databases was conducted from inception to July 15th, 2013. Crude odd ration (OR) with 95% confidence intervals (CI) was calculated. RESULTS: Thirteen case-control studies were assessed, including 2,019 oral cancer patients and 3,282 healthy controls. Meta-analysis showed that the overall association between GSTP1 Ile105Val polymorphism and susceptibility to oral cancer was not significant. However, further stratified analyses based on ethnicity indicated that this association was significant in East Asians (Val vs. Ile: OR=1.42, 95%CI: 1.06-1.90, P=0.019; Val/Val+Ile/Val vs. Ile/Ile: OR=1.59, 95%CI: 1.13-2.23, P=0.007) and Euro Caucasians (Val vs. Ile: OR=1.34, 95%CI: 1.03-1.75, P=0.032; Val/Val+Ile/Val vs. Ile/Ile: OR=1.66, 95%CI: 1.15-2.38, P=0.006), but not significant in South Asians and America Caucasians. CONCLUSIONS: This meta-analysis provides further evidence that GSTP1 Ile105Val polymorphism may be associated with oral cancer risk among East Asians and Euro Caucasians. However, further large-scale studies are needed to determine the association of GSTP1 Ile105Val genetic polymorphism with susceptibility to oral cancer.
