3种树脂水门汀对人牙龈成纤维细胞的体外细胞毒性

Cytotoxicity of 3 resin cements to human gingival fibroblasts

摘要: 目的: 比较3种不同树脂水门汀对人牙龈成纤维细胞(human gingival fibroblasts, HGFs)的体外细胞毒性。方法: 采用CCK-8法检测3种树脂水门汀(Panavia F、RelyXTM Unicem和Multilink Speed)48 h不同浓度(50%和100%)浸提液培养HGFs(ATCC,美国)24 h、72 h和120 h后的细胞相对增殖率(relative growth rate, RGR),判断其细胞毒性。采用SPSS 20.0 软件包分析不同树脂水门汀对HGFs的RGR是否存在显著影响。结果: 3种树脂水门汀不同浓度(50%和100%)浸提液培养HGFs的RGR为60.25%~100.02%,细胞毒性分级为0~2级,各组之间差异无显著性(P>0.05);固化过程中未涂布防氧化剂的Panavia F浸提液培养HGFs的RGR为10.67%~66.78%,细胞毒性分级为2~4级,与其他实验组相比,有显著差异(P<0.05)。结论: 3种树脂水门汀对体外培养的HGFs无细胞毒性,固化过程中未涂布防氧化剂的Panavia F对体外培养的HGFs有中度细胞毒性。

Abstract: PURPOSE: To study the cytotoxicity of 3 resin cements to human gingival fibroblasts (HGFs). METHODS: Three resin cements (Panavia F, RelyXTM Unicem and Multilink Speed) test samples were immersed and incubated in the culture medium for 48 h at 37℃. Cultured HGFs were exposed to two concentrations (50% and 100%) of material elutes for 24 h, 72 h and 120 h. The proliferation rate was evaluated using CCK-8 assay. The data were statistically analyzed by one-way analysis of variance using SPSS 20.0 software package. RESULTS: Relative growth rate(RGR) of all experimental groups ranged from 10.67% to 100.02%, the cytotoxicity grade of all groups was 0 to 4. There was no significant difference in the RGR among 3 resin cements, but the experimental group of Panavia F (uncovered with antioxidant) showed significantly lower RGR than other experimental groups. CONCLUSIONS: Panavia F, RelyXTM Unicem and Multilink Speed exhibit no cytotoxicity to HGFs, Panavia F(uncovered with antioxidant) shows moderate cytotoxicity.
