药物干预下骨质疏松大鼠正畸牙移动中TGF-β1表达的变化

Changes of TGF-β1 expression during orthodontic tooth movement in rats with osteoporosis

摘要: 目的 观察雷尼酸锶(strontium ranelate)、强骨胶囊(qianggu capsule)干预下骨质疏松(osteoporosis)大鼠正畸牙移动后牙周组织中转化生长因子β1(transforming growth factor beta 1,TGF-β1)的表达和分布,探讨2种药物的疗效。方法 选取60只3个月龄SD 雄性大鼠,随机分为实验对照组、模型对照组、雷尼酸锶组和强骨胶囊组,每组各15只。给予模型对照组、雷尼酸锶组、强骨胶囊组维A酸连续灌胃2周,行骨密度检测,确定骨质疏松模型成功建立。为所有大鼠安装正畸装置,分别在加力后7、14、21 d分批处死, 取大鼠上颌第一磨牙及附近牙槽骨组织块,进行H-E染色和免疫组织化学染色,半定量分析其牙周组织中TGF-β1的表达。采用SPSS 17.0 软件包对数据进行统计学分析。结果 雷尼酸锶组和强骨胶囊组TGF-β1表达较模型对照组显著增加(P<0.05);雷尼酸锶组TGF-β1表达显著强于强骨胶囊组(P<0.05)。结论 雷尼酸锶与强骨胶囊均可增强TGF-β1的表达,对骨质疏松大鼠骨代谢有正向调节作用,有利于牙的健康移动;雷尼酸锶的疗效强于强骨胶囊。

Abstract: ] PURPOSES: To observe the expression and distribution of TGF-β1 in periodontal tissue under intervention of Strontium ranelate and Qianggu capsule during orthodontic tooth movement in rats, and explore the efficacy of the 2 drugs. METHODS: Seventy male SD rats of 3 months old were selected in the study, and randomly divided into control group, model control group, Strontium ranelate group, Qianggu capsule group, each group had 15 animals. Retinoic acid was given by gavage to animals in the control group, Strontium ranelate group, Qianggu capsule group for 2 weeks, and bone density was detected to determine successful establishment of osteoporosis model. All rats were installed orthodontic device, and were sacrificed at 7 days, 14 days and 21 days, respectively. The tissue blocks of the first maxillary molar and adjacent alveolar bone were taken for H-E staining, immunohistochemical staining and semi-quantitative analysis was used to detect TGF-β1 expression in periodontal tissues. The data were statistically analyzed by SPSS 17.0 software package. RESULTS: TGF- beta 1 expression was significantly increased in Strontium ranelate group and Qianggu capsule group compared with control group (P<0.05); TGF- beta 1 expression in Strontium ranelate group was significantly stronger than that of Qianggu capsule group (P<0.05). CONCLUSIONS: Strontium ranelate and Qianggu capsule could enhance the expression of TGF- beta 1 and promote bone metabolism in osteoporosis rats, which is helpful to the movement of healthy teeth; the effect of Strontium ranelate is stronger than Qianggu capsule.
